Combination of the first and second step of SD protocol was investigated by using 100ppM BTB solution  as a sample, and the assay protocol from previous page, where Sv was set to 10 microliters Dv was varied from 0 to 80 microliters and Cv and D2v were set to zero. From the recording (A) and plotted data (B) follows that sample dilution of up to 10 times is obtained within 30 seconds. This two step protocol is useful for automation of the reagent based assays, since an appropriate reagent can be used in lieu of the diluent. Example is glucose assay discussed in 2.2.11.C.
